McKenna proposed in Food of the Gods (1992) that psilocybin mushrooms in the diet of early hominids accelerated the development of language and cognition. It remains a fringe idea with no supporting fossil or genetic evidence.

The Colorado River toad, also called the Sonoran Desert toad, secretes 5-MeO-DMT from its parotoid glands. Licking the animal is a persistent myth and a dangerous one, since the raw secretion also contains toxins.

Aldous Huxley (1894–1963) was a British writer and intellectual best known for the dystopian novel Brave New World (1932).In later life he became deeply interested in altered states of consciousness. His 1954 essay The Doors of Perception describes his experiences with mescaline and helped introduce psychedelic ideas to a wide Western audience. He continued to explore and write about psychedelics, mysticism, and human potential until his death.

Ecstasy (and Molly) are common street names for MDMA (3,4-methylenedioxymethamphetamine), a synthetic psychoactive substance. It produces stimulant effects (increased energy, alertness) combined with strong empathogenic or “entactogenic” effects—heightened feelings of emotional closeness, empathy, and well-being—along with mild psychedelic sensory changes. “Ecstasy” typically refers to pressed tablets (which may contain other substances), while “Molly” usually refers to the crystalline or powder form. MDMA is a Schedule I controlled substance in the United States and is illegal for recreational use in most countries.

Yopo (or Cohoba) is a traditional hallucinogenic snuff prepared from the seeds of the South American and Caribbean tree Anadenanthera peregrina (and closely related species). It was used for centuries—possibly millennia—by Indigenous peoples of the Orinoco basin, the Caribbean Taíno, and other groups in ritual, divinatory, and ceremonial contexts. Early European accounts, including those linked to Columbus’s voyages, describe its use. The seeds contain psychoactive tryptamines, principally bufotenine (5-HO-DMT), along with smaller amounts of DMT and 5-MeO-DMT, which produce intense, short-lasting visionary effects when insufflated.

Ibogaine is a psychoactive alkaloid extracted from the root bark of the West African plant Tabernanthe iboga. Observational and open-label studies (mostly conducted in clinics outside the United States) suggest that a single dose can rapidly reduce or suppress acute opioid withdrawal symptoms and temporarily lower drug craving in people with opioid use disorder. Some reports also indicate possible benefits for cocaine and certain other substance dependencies, though the evidence is thinner. These findings are limited: no large, rigorous randomized controlled trials have yet confirmed long-term efficacy, and most available data come from uncontrolled or retrospective observations. Significant safety risks exist, particularly cardiac effects such as QT-interval prolongation that can lead to arrhythmias. Fatalities have been reported, especially in unregulated settings. In the United States, ibogaine remains a Schedule I controlled substance and is not an approved medical treatment. Research, including studies of its metabolite noribogaine, continues under strict regulatory oversight.

“Businessman’s trip” is an old slang nickname for a DMT experience. Because the intense psychedelic effects of smoked or injected DMT typically last only 5–15 minutes, the term suggests that a person could take it during a short break and still return to work afterward—hence the joking comparison to a businessman squeezing in a quick “trip.”

Psychedelic tryptamines are a class of compounds built on the tryptamine molecular skeleton (an indole ring attached to an ethylamine side chain). Many of them act as agonists at the serotonin 5-HT2A receptor and produce classic psychedelic effects—altered perception, intense visual imagery, changes in thought and emotion, and shifts in the sense of self. Well-known examples include DMT, psilocin (from magic mushrooms), and 5-MeO-DMT.

DMT and monoamine oxidase (MAO) DMT is rapidly broken down by monoamine oxidase (MAO) enzymes, particularly MAO-A, which are abundant in the gut and liver. As a result, when DMT is taken orally on its own it is almost completely metabolized before it can reach the brain, producing little or no psychoactive effect. This rapid enzymatic degradation is the main reason pure oral DMT is inactive, while combinations that inhibit MAO (as in traditional ayahuasca) allow it to become active.

Rick Strassman is an American psychiatrist and clinical researcher best known for conducting the first government-approved human studies of DMT in the United States in over 20 years. Between 1990 and 1995 at the University of New Mexico, he administered approximately 400 doses of DMT to nearly 60 volunteers under controlled clinical conditions. His findings and reflections are detailed in his widely read 2001 book DMT: The Spirit Molecule. He is currently a clinical associate professor of psychiatry at the University of New Mexico School of Medicine.

The 5-HT2A receptor is a subtype of serotonin (5-hydroxytryptamine) receptor found abundantly in the brain’s cortex.DMT acts as a partial agonist at this receptor. When it binds and activates 5-HT2A receptors (especially on cortical pyramidal neurons), it disrupts normal sensory processing and network dynamics. This interaction is widely regarded as the primary mechanism underlying DMT’s intense psychedelic effects, similar to how classic psychedelics such as LSD and psilocybin also work primarily through 5-HT2A activation.

In 1931, German-Canadian chemist Richard Helmuth Fredrick Manske became the first person to synthesize DMT in a laboratory. He prepared and described DMT as part of a broader study of methyltryptamine derivatives, publishing the work in the Canadian Journal of Research. At the time it was treated as an ordinary chemical product with no known biological activity. Its presence in plants was not confirmed until 1946, and its intense hallucinogenic effects in humans were not discovered until 1956.

N,N-Dimethyltryptamine (DMT) is a naturally occurring tryptamine alkaloid and potent psychedelic compound.It is found in many plants (such as those used in ayahuasca) and animals, and is also produced endogenously in trace amounts in the human body. Chemically, it is a structural analog of the neurotransmitter serotonin. When consumed (typically via inhalation or in combination with MAO inhibitors), it produces intense, short-duration alterations in perception, cognition, and consciousness.

Current psychedelic research often looks at how compounds like LSD, psilocybin, DMT, mescaline, and ayahuasca affect brain connectivity and network activity.

Mescaline is the classic psychedelic associated with cactus species such as peyote and San Pedro. NIDA lists mescaline among classic psychedelics.

Ayahuasca combines DMT-containing plants with MAOI-containing plants, allowing DMT to remain active when taken orally.

Psilocybin is converted mainly into psilocin, which is the compound responsible for most of its psychedelic activity.
